• Formulation base upon that of the Alfred Hospital Department of Anatomical Pathology modified from Schmorl and the Armed Forces Institute of Pathology, substituting Isotonic Sodium Chloride for Sodium Citrate (which was orginally intended as an anticoagulant to allow "cleaner trephines); being intended for routine use on all Bone Marrow Trephines, and most small or prosected bone samples.
• Loss of staining properties especially nuclei, is minimal with this agent with over exposure of one or more days not causing serious defects. Recommended application time for trephines is 4-6 hrs is usually adequate, and in urgent situations material should be placed directly into fixative to simultaneously fix and decalcify and then processed. (one can often get away with 2-3 hours).
• QA assessment is by Certificate of Assessment protocal and requires validation and certification of all worksheets by at least two persons against all materials used and the Formulation Method.
